Cytogenetic analysis and chromosomal characteristics of the polymorphic 18S rDNA in the fish Prochilodus lineatus (Characiformes, Prochilodontidae) Genet. Mol. Biol.
Vicari,Marcelo Ricardo; Almeida,Mara Cristina de; Bertollo,Luiz Antonio Carlos; Moreira-Filho,Orlando; Artoni,Roberto Ferreira.
We used differential staining techniques (BSG, GTG, AgNO3, DAPI and CMA3 banding) and fluorescent in situ hybridization (FISH) with 5S and 18S probes to investigated the karyotypic and cytogenetic chracteristics of Prochilodus lineatus specimens from a population in Vila Velha state park (Parque Estadual de Vila Velha, Ponta Grossa, Paraná state, southern Brazil). The specimens studied showed the same karyotype as that found in other P. lineatus populations, i.e. 2n = 54 biarmed chromosomes (40m + 14 sm) and c-positive heterochromatin preferentially located pericentromerically in all chromosomes. The presence of partial or totally heterochromatic supernumerary chromosomes with numeric intra-individual variation was confirmed in the analyzed population. The...

Description of novel microsatellite loci in the Neotropical fish Prochilodus argenteus and cross-amplification in P. costatus and P. lineatus Genet. Mol. Biol.
Barbosa,Anna C.D.R.; Galzerani,Felipe; Corrêa,Thaís C.; Galetti Jr.,Pedro M.; Hatanaka,Terumi.
Prochilodus is one of the most important fish resources of South America, in addition to the important role it plays in nutrient cycling of Neotropical rivers. In the present study, we describe the isolation and characterization of nine novel microsatellite loci in Prochilodus argenteus. The number of alleles per polymorphic locus varied from 5 (Par76) to 21 (Par85), revealing a total of 116 alleles. The values of observed and expected heterozygosities ranged from 0.629 (Par69) to 0.926 (Par85 and Par86) and from 0.643 (Par66) to 0.931 (Par80), respectively. Furthermore, the ability of these and other previously described microsatellite markers to amplify orthologous loci was tested in two related species, Prochilodus costatus and Prochilodus lineatus....

Development of the digestive system in larvae of the Neotropical fish Prochilodus argenteus (Characiformes, Prochilodontidae) Animal Sciences
Santos, Alcione Eneida; Pedreira, Marcelo Mattos; Santos, Thais Garcia; Moura, Guilherme de Souza; Santos, José Claudio Epaminondas dos; Silva, Robson Campos.
The development of the digestive system in larvae of the Neotropical fish P. argenteus was analyzed histologically. On the 3rd day after hatching, the digestive system comprised oropharyngeal cavity, esophagus and simple undifferentiated tube. Since secreting cells, positive to Alcian Blue (AB), were found in the esophagus, digestive activity in the initial phase had occurred. On the 18 and 28th days after hatching, the esophagus was positive for AB and Periodic Acid Schiff (PAS) stain. The stomach was fully differentiated, with the cardiac, fundic and pyloric regions. Different regions of the epithelium were characterized by basic and acidic secreting cells (AB and PAS positive). On the 18 and 28th days after hatching, the intestine was long, coiled and...

Effects of Biodegradable Detergents in the Accumulation of Lipofuscin (Age Pigment) in Gill and Liver of Two Neotropical Fish Species International Journal of Morphology
Pereira,Bruno Fiorelini; Alves,Anderson Luis; Senhorini,José Algusto; Rocha,Rita de Cássia Gimenes de Alcântara; Pitol,Dimítrius Leonardo; Caetano,Flávio Henrique.
Currently there is a growing concern, in both population and governments, to identify the effects of substances commonly disposed of into rivers and lakes, on aquatic fauna and flora. Thus the objective of the present study was to verify effects of biodegradable detergents and water from an urban lake on gills and liver of two neotropical fish species of great economic importance, Astyanax altiparanae and Prochilodus lineatus. In order to do so, lipofuscin, also called the ageing pigment, was used as bioindicator. After one and five months of experiment both tissues accumulated this pigment. These data are discussed from physiological points of view, related with lipid peroxidation and mitochondrial damage.

RAPD markers indicate the occurrence of structured populations in a migratory freshwater fish species Genet. Mol. Biol.
Hatanaka,Terumi; Galetti Jr.,Pedro Manoel.
Many factors have contributed to the destruction of fish habitats. Hydroelectric dams, water pollution and other environmental changes have resulted in the eradication of natural stocks. The aim of this study was to detect the genetic variation in Prochilodus marggravii from three collection sites in the area of influence of the Três Marias dam (MG) on the São Francisco river (Brazil), using the RAPD technique. The results obtained revealed that the fish in the downstream region nearest the dam have a higher similarity coefficient than those from the other sampling sites that may be related to differences in environmental characteristics in these regions. Additionaly, significant differences in the band frequencies were observed from one collection site to...

Threatened fish spawning area revealed by specific metabarcoding identification of eggs and larvae in the Beni River, upper Amazon ArchiMer
Miranda-chumacero, Guido; Mariac, Cédric; Duponchelle, Fabrice; Painter, Lilian; Wallace, Robert; Cochonneau, Gérard; Molina-rodriguez, Jorge; Garcia-davila, Carmen; Renno, Jean-françois.
Identifying fish spawning areas is of great ecological and conservation importance as fishes are suffering increasing threats levels from anthropogenic activities. However, to date very few studies have done so in the Amazon basin. In the Beni River located in the upper Madeira basin, fishers reported that a particular ecotone near Rurrenabaque city was a fish reproduction area. To test the importance of this zone as a spawning site, we conducted an ichthyoplankton survey during the month when reproduction is most likely to occur. The specific identification of larvae and eggs was made with a metabarcoding analysis. With this approach 13 different fish species of high importance for regional and local fisheries were identified, including the long-distance...
